Background

Protein phosphorylation, which dynamically regulates protein functions, localization, and interactions, mediates numerous cellular processes. Detecting and characterizing phosphorylation states requires protection from endogenous phosphatases that reverse phosphorylation during sample preparation. The Phosphatase Inhibitor Cocktail is composed of two distinct inhibitor formulations: Tube A, provided in DMSO, targets serine/threonine protein phosphatases (PP1, PP2A isoforms) and alkaline phosphatase isoenzymes through inhibitors such as Cantharidin, Bromotetramisole, and Microcystin LR; Tube B, in aqueous solution, inhibits tyrosine phosphatases, acid and alkaline phosphatases using Sodium orthovanadate, Sodium molybdate, Sodium tartrate, Imidazole, and Sodium Fluoride. This cocktail enables researchers to maintain phosphorylation integrity during cell lysis and sample processing for downstream biochemical assays, including immunoblotting, immunoprecipitation, kinase activity analyses, and mass spectrometry.

Protocol

Thawed at room temperature, then diluted to solution samples (such as cell lysates or tissue extracts) at 1:100 (v/v) before the experiment, add A tube and B tube successively: add A tube, mix well; Add B tube, mix well (do not mix A tube and B tube and then add together).
