Diphenyleneiodonium chloride (DPI, CAS 4673-26-1) is an agonist of G protein-coupled receptor 3 (GPR3), a Gs-linked GPCR that promotes intracellular cAMP accumulation. In GPR3-expressing HEK293 cells, DPI elevates cAMP levels independently from its known inhibitory actions on NADH oxidases (NOX). DPI also induces receptor desensitization, calcium influx, and β-arrestin2 recruitment in HeLa cells transfected with GPR3. Additionally, DPI potently inhibits NOX activity (EC50=0.1 μM), nitric oxide synthase irreversibly, and cytochrome P450 reductase (Ki=2.8 μM). DPI is broadly utilized for probing cAMP-related signaling and redox enzyme functions.
