WZB117
WZB117 (CAS 1223397-11-2) is a small-molecule inhibitor that targets glucose transporter 1 (GLUT1), a membrane protein responsible for basal glucose uptake essential to cellular bioenergetics. WZB117 selectively suppresses GLUT1-mediated glucose transport, exhibiting an IC50 of approximately 0.6 μM in various cancer cell lines. In vitro studies demonstrate that exposure to WZB117 reduces GLUT1 protein levels, intracellular ATP content, and expression of glycolytic enzymes, triggering activation of AMP-activated protein kinase (AMPK), downregulation of cyclin E2, reduced phosphorylated retinoblastoma, cell cycle arrest, senescence, and cell death by necrosis. In vivo, administration of WZB117 significantly decreases tumor growth in A549 lung cancer xenograft models, indicating its potential utility as a research tool in cancer metabolism studies.
