Background

HPF (CAS 359010-69-8), hydroxyphenyl fluorescein, is a cell-permeable aromatic aminofluorescein derivative exhibiting minimal intrinsic fluorescence. Upon oxidation by highly reactive oxygen species (hROS), such as hydroxyl radicals, peroxynitrite, or those enzymatically generated via peroxidase/H?O? systems, HPF is converted into fluorescein, emitting strong green fluorescence (excitation/emission maxima: 490/515 nm). Notably, HPF does not respond to hypochlorite, nitric oxide, hydrogen peroxide, or superoxide ions, demonstrating specificity in detecting hROS. It is suitable for visualizing intracellular oxidative stress using fluorescence microscopy, microplate readers, high-throughput imaging systems, and flow cytometry.
