Background

Tauroursodeoxycholic Acid (CAS No.: 14605-22-2) is a taurine-conjugated derivative of the secondary bile acid ursodeoxycholic acid and is widely utilized in biomedical research as a cytoprotective agent with prominent anti-apoptotic and endoplasmic reticulum (ER) stress–modulating properties. Functionally, it acts as a chemical chaperone that stabilizes protein folding and mitigates ER stress signaling by reducing activation of ER stress-associated proteins and preventing dissociation of key regulatory complexes such as GRP78 and PERK, thereby attenuating unfolded protein response pathways. TUDCA has also been shown to suppress apoptosis through downregulation of caspase-3 and caspase-12 activation and modulation of ERK and Akt-dependent signaling pathways, while contributing to mitochondrial stabilization. In cellular systems, it demonstrates protective effects across a range of stress-related conditions, including oxidative stress and protein misfolding, typically exhibiting activity in the low micromolar range depending on assay conditions. In vivo, TUDCA has been reported to improve tissue perfusion, enhance angiogenesis, and increase survival of transplanted mesenchymal stem cells in ischemic models, as well as to exert neuroprotective effects in models of neurodegenerative diseases and retinal injury. Owing to these properties, it is frequently employed in studies of ER stress-related pathologies, neurodegeneration, metabolic disorders, and regenerative medicine, as well as in drug discovery efforts targeting apoptosis and protein misfolding pathways, with experimental concentrations or dosing regimens varying according to specific model systems and research objectives.
