Background

Isoallolithocholic acid is a stereoisomeric derivative of Lithocholic acid and belongs to secondary bile acids. As endogenous metabolites, Isoallolithocholic acid exhibits unique functions in immune regulation: By activating nuclear receptors (such as fanitol X receptor FXR or retinoic acid-related orphan receptor RORγt), it specifically promotes the differentiation and function of regulatory T cells (Tregs), enhances their immunosuppressive activity (such as the secretion of IL-10 and TGF-β), and thereby inhibits excessive inflammatory responses. Studies have shown that Isoallolithocholic acid can induce naive T cells to differentiate into Tregs by regulating intracellular signaling pathways (such as AKT/mTOR) or epigenetic modifications (such as histone acetylation) within T cells. It has potential therapeutic value in models of autoimmune diseases such as colitis and multiple sclerosis.
