EN460 is a selective endoplasmic reticulum oxidoreductase 1 (ERO1) inhibitor that specifically binds to the reduced conformation of ERO1, blocking the enzyme’s oxidative reaction process and inhibiting its re-oxidation. ERO1 participates in oxidative protein folding in the endoplasmic reticulum by catalyzing protein disulfide bond formation. By inhibiting ERO1 activity during this oxidation process, EN460 regulates redox homeostasis in the endoplasmic reticulum and is widely used to study mechanisms of protein folding, endoplasmic reticulum stress, and related diseases.
