2-Thio-UTP is a base-modified ribonucleoside triphosphate commonly used in RNA-based biomedical research. Incorporation of 2-Thio-UTP into RNA transcripts reduces innate immune activation compared to unmodified RNA. Mechanistically, RNAs transcribed with 2-Thio-UTP lack the ability to activate RNA-dependent protein kinase (PKR), markedly diminish 2'-5'-oligoadenylate synthetase (OAS) pathway induction, decrease recognition by retinoic acid-inducible gene I (RIG-I), and suppress Toll-like receptor (TLR) mediated signaling. Thus, 2-Thio-UTP serves as a research tool in studies designing low-immunogenic RNAs for therapeutic applications.
