Zoxazolamine
Zoxazolamine (CAS No.: 61-80-3) is a centrally acting skeletal muscle relaxant historically utilized as an antispasmodic and uricosuric agent, and is now predominantly employed as a pharmacological probe in biomedical research. It is extensively metabolized by multiple cytochrome P450 isoforms, making it a well-established functional substrate for assessing hepatic microsomal enzyme activity; in particular, the duration of zoxazolamine-induced paralysis in rodent models serves as a sensitive in vivo indicator of alterations in CYP450 expression and metabolic capacity. Through its central myorelaxant action, which is associated with modulation of neuronal excitability and neuromuscular transmission, zoxazolamine provides a useful tool for investigating neuropharmacological mechanisms and xenobiotic metabolism. In addition to its metabolic applications, it has been shown to influence cellular signaling in skin-related systems, including dose-dependent modulation of pigmentation-associated factors in keratinocyte–melanocyte co-culture models, suggesting broader utility in studies of cellular differentiation and intercellular communication. Its biological activity is typically observed in the low micromolar to micromolar range depending on the assay system, and experimental concentrations or dosing regimens vary according to specific study designs. Overall, zoxazolamine remains a valuable compound for drug metabolism studies, enzyme induction or inhibition screening, and mechanistic investigations in both in vitro and in vivo models.
