Zinc protoporphyrin IX
Zinc protoporphyrin IX (CAS No.: 15442-64-5) is an endogenous heme analog generated during heme biosynthesis under conditions of iron deficiency or impaired iron utilization, and is widely utilized in biomedical research as a functional probe of heme metabolism and oxidative stress pathways. It acts primarily as a competitive inhibitor of heme oxygenase-1 (HO-1), the rate-limiting enzyme in heme degradation, thereby modulating the production of biologically active metabolites such as bilirubin and carbon monoxide, and influencing cellular redox homeostasis and cytoprotective signaling. Zinc protoporphyrin IX has been shown to regulate HO-1 expression at the transcriptional level, with context-dependent effects that may either induce or suppress enzyme expression depending on cell type and experimental conditions, and it can interfere with Nrf2/HO-1 signaling pathways involved in antioxidant defense. In cellular systems, it attenuates the protective effects of antioxidant compounds against hydrogen peroxide-induced oxidative damage, including reductions in reactive oxygen species scavenging and increased susceptibility to DNA damage. Beyond HO-1 inhibition, it exhibits additional biological activities, including inhibition of soluble guanylyl cyclase and nitric oxide synthase isoforms at low micromolar concentrations, supporting its role as a modulator of gasotransmitter signaling. This compound is commonly applied as a biomarker for iron status and in studies of iron metabolism disorders, as well as in oncology and inflammation research to investigate the role of heme oxygenase pathways, with experimental concentrations varying according to specific in vitro or in vivo study designs.
