Triparanol
Triparanol (CAS No.: 78-41-1) is a synthetic small-molecule inhibitor of cholesterol biosynthesis that primarily targets 24-dehydrocholesterol reductase (24-DHCR), thereby blocking the conversion of sterol intermediates into cholesterol and leading to reduced intracellular cholesterol levels. By perturbing sterol homeostasis, Triparanol also modulates Hedgehog (Hh) signaling, a pathway critically dependent on sterol-mediated posttranslational processing; it interferes with the maturation of Hh ligands and disrupts the sterol-sensing domain of the receptor PTCH1, resulting in attenuation of downstream pathway activity. This dual impact on cholesterol metabolism and Hh signaling has made Triparanol a valuable experimental tool for investigating the interplay between lipid metabolism and developmental signaling pathways, particularly in studies of tumor cell proliferation and pathway-driven oncogenic processes. In vitro studies indicate that its biological effects are typically observed in the low micromolar range, although experimental conditions may vary depending on cell type and assay design. Triparanol is widely applied in cell-based assays and preclinical models to explore sterol-dependent signaling mechanisms, to dissect regulatory nodes within the Hh pathway, and to support compound screening efforts targeting metabolic and signaling crosstalk; dosing and exposure parameters are generally optimized according to specific experimental objectives.
