Background

Tentoxin (CAS No.: 28540-82-1) is a cyclic tetrapeptide mycotoxin produced by Alternaria species that is widely employed as a biochemical probe in plant and chloroplast research. It functions primarily as a selective inhibitor of the chloroplast ATP synthase complex, targeting the β subunit and disrupting photophosphorylation and energy transfer processes essential for photosynthetic activity. This mechanism leads to impaired chloroplast development and characteristic chlorosis in susceptible plant tissues, reflecting its role as a selective phytotoxin and herbicidal agent. In vitro studies indicate that Tentoxin exerts inhibitory activity on chloroplast ATP synthesis in the low micromolar range, although potency may vary depending on species and experimental conditions. Owing to its well-defined mode of action, Tentoxin is extensively utilized to investigate the structure–function relationships of ATP synthase, the regulation of photosynthetic energy conversion, and stress-related signaling pathways in plant systems. It also serves as a valuable tool in studies of chloroplast biogenesis, protein expression, and metabolic adaptation, as well as in screening platforms aimed at identifying modulators of energy metabolism. Experimental concentrations and treatment regimens are typically optimized according to specific cellular or physiological models to achieve controlled perturbation of chloroplast function.
