Background

TCPOBOP (CAS No.: 76150-91-9) is a potent and selective agonist of the constitutive androstane receptor (CAR), widely utilized as a chemical probe in hepatic biology and xenobiotic metabolism research. It exhibits strong species specificity, robustly activating mouse CAR at nanomolar potency while showing minimal activity toward human or rat CAR, making it particularly valuable in murine models. Upon CAR activation, TCPOBOP drives transcriptional programs that induce cytochrome P450 monooxygenases, multidrug resistance proteins, and xenobiotic efflux transporters, thereby modulating hepatic detoxification pathways and metabolic homeostasis. Functionally, it promotes pronounced hepatocyte proliferation and liver enlargement without overt tissue injury, and has been shown to attenuate Fas-mediated liver damage in mice through regulation of apoptosis-associated proteins such as Bcl-2.
