TBK1/IKKε-IN-5
TBK1/IKKε-IN-5 (CAS No.: 1893397-65-3) is a potent, orally active dual inhibitor of TANK-binding kinase 1 (TBK1) and IκB kinase ε (IKKε), two noncanonical IκB kinases that play central roles in innate immune signaling, interferon regulation, and tumor-associated inflammatory pathways. This small-molecule inhibitor exhibits high potency in the nanomolar range, with reported inhibitory activity against TBK1 and IKKε at low nanomolar concentrations, enabling effective modulation of downstream signaling cascades such as IRF3/7 activation and cytokine production. In cellular models, TBK1/IKKε-IN-5 has been shown to suppress the production of immunosuppressive cytokines in tumor spheroid systems without inducing cytotoxicity, while enhancing T cell activation markers, including increased secretion of IL-2 and IFN-γ in primary human CD4+ and CD8+ T cells as well as in Jurkat T cell models. In ex vivo organotypic tumor spheroid platforms characterized by immunosuppressive microenvironments, inhibition of TBK1/IKKε signaling has been associated with improved responsiveness to immune checkpoint blockade, providing a mechanistic basis for combination strategies in tumor immunology research. In syngeneic mouse tumor models, co-administration with PD-L1 blockade has demonstrated enhanced tumor growth control and prolonged survival compared to single-agent conditions, supporting its utility as a tool compound for investigating kinase-driven immune regulation and resistance mechanisms. TBK1/IKKε-IN-5 is therefore widely applied in studies of innate immunity, cancer immunobiology, and kinase-targeted drug discovery, with experimental concentrations or dosing regimens determined by specific assay systems and research objectives.
