Background

SRI-37330 (CAS No.: 2322245-42-9) is an orally bioavailable small-molecule inhibitor of thioredoxin-interacting protein (TXNIP), a key regulator of cellular redox balance and metabolic homeostasis, widely studied in the context of metabolic disorders and endocrine signaling. By suppressing TXNIP expression at both mRNA and protein levels in cellular systems, SRI-37330 modulates downstream pathways involved in glucose and lipid metabolism, including the regulation of pancreatic α-cell function and glucagon secretion, as well as hepatic gluconeogenesis. This dual action results in reduced hepatic glucose output and improvement of lipid handling, contributing to attenuation of hepatic steatosis. In vitro studies demonstrate activity in the low micromolar range, along with the ability to inhibit inflammatory cell death processes such as pyroptosis under specific experimental conditions.
