SEL120-34A hydrochloride
SEL120-34A hydrochloride (CAS No.: 1609452-30-3) is a highly selective ATP-competitive inhibitor of CDK8/CDK19.
In vitro, it exhibits IC₅₀ values of approximately 4.4 nM and 10.4 nM against CDK8/CycC and CDK19/CycC, respectively. It shows weak inhibition against other CDKs (1/2/4/5/6/7) and has a selectivity index of >200-fold over CDK9.
In cells, SEL120-34A inhibits the serine phosphorylation of STAT1-S727 and STAT5-S726, thereby affecting transcription elongation and the expression of inflammation- and tumor-associated genes.
SEL120-34A demonstrates significant anti-proliferative activity in a subset of AML cell lines with high STAT1/5 phosphorylation. It suppresses tumor growth, reduces leukemia burden, and shows favorable tolerability in acute myeloid leukemia xenograft models.
Accordingly, it is used as a representative molecule for targeted agents against the CDK8/CDK19-STAT axis in mechanistic and pharmacological studies.
