S2157, an N-alkylated trans-cyclopropylamine (TCP) derivative, is a potent inhibitor of lysine-specific demethylase 1.S2157 increases H3K9 methylation and corresponding H3K27 deacetylation in the super-enhancer region.S2157 inhibits transcription of the NOTCH3 and TAL1 genes, which induces TCP-resistant acute lymphoblastic leukemia T- ALL cells apoptosis.S2157 effectively crosses the blood-brain barrier and almost completely eradicates CNS leukemia in T-ALL cell transplanted mice.
