S2116, an N-alkylated trans-cyclopropylamine (TCP) derivative, is a potent inhibitor of lysine-specific demethylase 1. S2116 increases H3K9 methylation and corresponding H3K27 deacetylation in the super-enhancer region.S2116 inhibits transcription of the NOTCH3 and TAL1 genes, which induces TCP-resistant acute lymphoblastic leukemia T- ALL cell apoptosis.S2116 significantly delayed the growth of T-ALL cells in xenograft mice.
