Roblitinib (CAS No.: 1708971-55-4) is a synthetic, small-molecule, orally available, and highly selective human fibroblast growth factor receptor 4 (FGFR4) tyrosine kinase inhibitor intended for applications such as tumor pathway biology and target validation in biomedical research and drug discovery settings; it directly inhibits receptor catalytic activity through reversible covalent binding to the FGFR4 kinase domain, thereby suppressing downstream signaling networks that regulate processes such as cell proliferation, differentiation, survival, and angiogenesis, and shows mechanism-consistent inhibitory effects in research models with elevated FGFR4 activity. Existing biochemical and cellular evidence indicates that its in vitro potency is in the nanomolar range, and that it produces significant inhibition of cell proliferation in FGFR4-dependent cellular models (such as Huh7); broad kinase profiling studies show that it has a pronounced selectivity advantage for FGFR4, approximately three orders of magnitude higher than most tested kinases. In preclinical research species, regulation of bile acid synthesis consistent with target engagement and related changes in circulating cholesterol can be observed, providing translatable pharmacodynamic readouts for in vivo mechanistic studies, without involving any clinical application.
