RCGD423
RCGD423 (CAS No.: 108237-91-8) is a small-molecule modulator of gp130-mediated signaling with demonstrated activity in regulating cartilage homeostasis and repair, making it a valuable tool compound in musculoskeletal and osteoarthritis research. Mechanistically, RCGD423 enhances gp130 downstream signaling, leading to increased phosphorylation of STAT3 and upregulation of Myc in articular chondrocytes, while its effects can be attenuated by gp130 pathway inhibitors, supporting target specificity. In vitro studies show that RCGD423 acts at low micromolar concentrations to suppress chondrocyte hypertrophy markers such as RUNX2 and COL10, reduce apoptosis and degeneration induced by pro-hypertrophic stimuli including BMP4, and promote chondrocyte proliferation in both healthy and osteoarthritic human cells. In vivo, it has been shown to reduce cartilage loss and improve tissue integrity in rodent models of joint injury, consistent with its role in preventing articular cartilage degeneration and promoting repair. RCGD423 is therefore widely applied in cellular and animal models to investigate gp130/STAT3 pathway modulation, chondrocyte biology, and regenerative strategies, with experimental concentrations or dosing regimens adjusted according to specific study designs and therapeutic hypotheses.
