Pyridoxamine dihydrochloride
Pyridoxamine dihydrochloride (CAS No.: 524-36-7) is a bioactive derivative of vitamin B6 that functions as an inhibitor of advanced glycation end products (AGEs) and advanced lipoxidation end products (ALEs), key contributors to oxidative stress and tissue damage in metabolic disorders. As a precursor to pyridoxal 5'-phosphate, it participates in cellular metabolism while exerting protective effects through the interception of reactive carbonyl intermediates, thereby disrupting protein glycation and lipid peroxidation pathways. Pyridoxamine dihydrochloride has been shown to scavenge reactive dicarbonyl species such as isoketals and inhibit the formation of toxic adducts, including lysyl-levuglandin-lactam complexes, supporting its role in mitigating oxidative and carbonyl stress. In experimental systems, it demonstrates activity in the micromolar to millimolar range in cell-free and cellular assays and has been widely employed in studies of diabetic complications, particularly in models of retinal vascular injury where it reduces pathological features such as acellular capillary formation and accumulation of glycation markers. Additionally, it has been used in animal models to investigate cognitive and vascular dysfunction associated with diabetes, highlighting its utility as a research tool for probing AGE- and ALE-mediated signaling pathways and for evaluating therapeutic strategies targeting metabolic and oxidative stress-related diseases.
