Background

NSC-87877 disodium (CAS No.: 56932-43-5) is a synthetic, cell membrane-permeable small-molecule inhibitor of protein tyrosine phosphatases, originally identified from a screen of the National Cancer Institute diversity compound library, and is commonly used in signal transduction research as well as oncology- and immunology-oriented early discovery programs; its main targets are the SH2 domain-containing phosphatases SHP?2 and SHP?1, and as a dual-target inhibitor it can modulate downstream signal transmission from receptor tyrosine kinases to the Ras–Raf–MEK–ERK axis, showing inhibitory activity in the nanomolar to submicromolar range in biochemical assays (typical IC50 approximately SHP?2 0.318 μM, SHP?1 0.355 μM), inhibiting epidermal growth factor-induced ERK1/2 activation in HEK293 cells in cellular systems, and reducing cell viability and proliferation in MDA?MB?468 breast-derived cells, with pathway engagement and phenotypic effects typically observed at low micromolar levels; in addition to its main targets, this compound can also inhibit the dual-specificity phosphatase DUSP26, therefore when assessing selectivity across the broader phosphatase family, orthogonal biochemical assays and genetic controls are recommended to ensure robust conclusions; typical applications include use as a reference inhibitor in phosphatase panels, pathway-dependency mapping and target validation in mammalian cell models, and as a comparator benchmark compound in early screening cascades, with the concentrations or doses used in experiments generally optimized according to the specific experimental design and research objectives; this product is suitable for basic and mechanistic research related to life sciences and drug discovery, and is for laboratory research use only.
