Background

NBI 31772 (CAS No.: 374620-70-9) is a non-peptidic, non-selective inhibitor of insulin-like growth factor binding proteins (IGFBPs) that functions by disrupting IGF–IGFBP interactions, thereby increasing the bioavailability of free, biologically active insulin-like growth factors, particularly IGF-1. It exhibits high affinity for multiple IGFBP subtypes, with inhibitory activity in the nanomolar range, enabling modulation of IGF-dependent signaling pathways implicated in cell survival, metabolism, and tissue regeneration. Mechanistically, NBI 31772 enhances IGF-mediated signaling by releasing sequestered ligands from IGFBPs, which has been associated with neuroprotective effects in experimental models of cerebral ischemia, including reduction of infarct volume, as well as promotion of anabolic processes such as proteoglycan synthesis in osteoarticular chondrocytes. Additionally, it has been shown to potentiate IGF3-driven biological responses in zebrafish oocyte maturation assays, supporting its role as a tool compound for studying IGF axis biology across species. In vitro and in vivo, NBI 31772 is widely utilized to investigate IGF-related mechanisms in neuronal survival, skeletal muscle regeneration, and metabolic regulation, with experimental concentrations or dosing regimens typically adjusted according to specific model systems and study objectives in biomedical research and drug discovery contexts.
