Naphthofluorescein

Naphthofluorescein (CAS No.: 61419-02-1) is a synthetic small-molecule probe and bioactive compound widely used in biomedical research for its dual functionality as an enzyme inhibitor and fluorescent reporter. It has been characterized as a cell-permeable inhibitor of the proprotein convertase furin, exhibiting activity in the low micromolar range, and consequently interfering with furin-mediated processing of substrates such as membrane type-1 matrix metalloproteinase, leading to reduced downstream activation of matrix metalloproteinase-2 and attenuation of cell motility and invasive behavior in tumor cell models. In parallel, Naphthofluorescein disrupts the interaction between hypoxia-inducible factor-1 and Mint3, thereby suppressing Mint3-dependent HIF-1 transcriptional activity and glycolytic reprogramming in cancer cells and macrophages without evident cytotoxicity in vitro or adverse effects in vivo under experimental conditions.
