Naphthofluorescein
Naphthofluorescein (CAS No.: 61419-02-1) is a synthetic small-molecule probe and bioactive compound widely used in biomedical research for its dual functionality as an enzyme inhibitor and fluorescent reporter. It has been characterized as a cell-permeable inhibitor of the proprotein convertase furin, exhibiting activity in the low micromolar range, and consequently interfering with furin-mediated processing of substrates such as membrane type-1 matrix metalloproteinase, leading to reduced downstream activation of matrix metalloproteinase-2 and attenuation of cell motility and invasive behavior in tumor cell models. In parallel, Naphthofluorescein disrupts the interaction between hypoxia-inducible factor-1 and Mint3, thereby suppressing Mint3-dependent HIF-1 transcriptional activity and glycolytic reprogramming in cancer cells and macrophages without evident cytotoxicity in vitro or adverse effects in vivo under experimental conditions. In addition to its inhibitory properties, it functions as a pH-sensitive fluorescent dye, enabling its application in functional imaging modalities, including Cerenkov-based approaches, and in studies of intracellular pH dynamics.
