Background

N-Octyl-β-D-thioglucopyranoside (CAS No.: 85618-21-9) is a low-molecular-weight, nonionic detergent widely employed in membrane biochemistry and structural biology for the gentle solubilization and stabilization of membrane-associated proteins. Functionally, it disrupts lipid–protein and protein–protein interactions without inducing significant denaturation, thereby preserving native protein conformation and activity in aqueous environments. This property makes it particularly valuable for extracting and purifying membrane proteins, including those from bacterial systems such as E. coli, as well as for downstream applications in proteomics, enzymology, and high-resolution structural studies such as crystallography. The compound exhibits a critical micelle concentration in the millimolar range and forms micelles that efficiently encapsulate hydrophobic domains, facilitating protein dispersion while maintaining functional integrity. It is stable in aqueous solutions, resistant to enzymatic degradation by β-glucosidases, and can be readily removed by dialysis, enabling compatibility with sensitive biochemical assays.
