ML-264
ML-264 (CAS 1550008-55-3) is a selective small molecule inhibitor targeting Krüppel-like factor 5 (KLF5), a transcription factor that facilitates proliferation in intestinal epithelial cells and is implicated in colorectal cancer progression. Mechanistically, ML-264 suppresses KLF5 expression and disrupts downstream MAPK signaling by reducing EGR1 levels, a transcriptional activator of KLF5. In cell-based assays, ML-264 shows potent antiproliferative activity against colorectal cancer lines (DLD-1 IC50: 29 nM; HT29 and SW620 IC50: 130 nM), sparing non-cancerous IEC-6 cells. In vivo, ML-264 inhibits tumor growth in colorectal cancer xenograft models, highlighting its relevance as a research tool in cancer biology.
