Methyclothiazide (CAS No.: 135-07-9) is a thiazide diuretic mainly used to treat cardiovascular-related diseases such as hypertension and edema. Its mechanism of action involves inhibiting the reabsorption of sodium and chloride ions in the renal tubules, thereby increasing urine output and reducing blood volume, which in turn lowers blood pressure; in addition, Methyclothiazide can also reduce vascular responses induced by endogenous vasoconstrictive stimuli (such as norepinephrine) by antagonizing voltage-dependent calcium channel (VDCC) activity, exhibiting an antihypertensive effect. In vitro studies show that Methyclothiazide can significantly inhibit VDCC activity in the low micromolar concentration range, leading to relaxation of vascular smooth muscle; in cell and animal models, Methyclothiazide is often used to evaluate its effects on vasoconstrictive responses, and the doses or concentrations used in experiments usually depend on the experimental design and research objectives.
