Lamotrigine is an effective oral anticonvulsant or antiepileptic agent that selectively blocks voltage-gated sodium channels, stabilizes presynaptic neuronal membranes, and inhibits glutamate release. In in vitro studies, lamotrigine blocks sustained repetitive neuronal firing in a dose-dependent manner and effectively inhibits the release of glutamate and aspartate, but has no significant effect on the release of acetylcholine and γ-aminobutyric acid. Lamotrigine is mainly inactivated via hepatic microsomal glucuronidation metabolism; its mechanism of action is similar to that of phenytoin and carbamazepine, producing a slight frequency-dependent inhibition of depolarizing potentials by modulating fast voltage-dependent sodium currents.
