Lactisole
Lactisole (CAS No.: 150436-68-3) is a small-molecule antagonist of sweet taste receptors that functions primarily by selectively targeting the T1R3 subunit of the heterodimeric T1R2/T1R3 receptor complex, a key mediator of sweet taste perception and glucose sensing. By binding to T1R3, Lactisole inhibits receptor activation induced by natural and artificial sweeteners, thereby attenuating downstream signal transduction pathways associated with sweetness perception and reducing both the intensity and persistence of sweet taste. In addition to its sensory effects, this compound has been widely utilized as a pharmacological tool to investigate the role of sweet taste receptors in metabolic regulation, including modulation of glucose-stimulated insulin secretion in pancreatic β-cells and hormone release such as glucagon-like peptides from enteroendocrine cells. Mechanistically, Lactisole also influences receptor crosstalk by enhancing inhibitory interactions between umami signaling and sweet taste pathways. Its activity is typically observed in the low micromolar range in in vitro systems, making it suitable for receptor function studies, ligand–receptor interaction analyses, and pathway validation experiments.
