Irinotecan HCl Trihydrate (CAS 136572-09-3), also known as CPT-11, is a semisynthetic derivative of camptothecin designed to improve therapeutic efficacy while reducing toxicity. It acts as a soluble prodrug, which undergoes enzymatic cleavage by carboxylesterases in vivo to yield SN-38, a potent inhibitor of topoisomerase I. CPT-11 exhibits cytotoxicity against leukemia (P388) cell lines with an IC50 of 3.4 μM, and demonstrates antitumor activity in vitro and in vivo against both drug-sensitive and multidrug-resistant tumor models. It is FDA-approved for treating metastatic colorectal cancer following progression after fluorouracil-based therapies.
