Background

IACS-010759 (CAS No.: 1570496-34-2) is a synthetic small-molecule mitochondrial oxidative phosphorylation inhibitor intended for biomedical research and early-stage drug discovery. As a selective Complex I (NADH dehydrogenase) modulator, it is used in mechanistic studies in the fields of cancer metabolism and mitochondrial bioenergetics; by inhibiting the ubiquinone-reducing activity of Complex I in the electron transport chain, it suppresses oxidative phosphorylation, reduces basal and spare respiratory capacity, and induces compensatory enhancement of glycolysis, thereby triggering cellular metabolic reprogramming under conditions dependent on mitochondrial metabolism. In biochemical and cellular systems, its direct action on Complex I is typically observed at nanomolar potency, while cellular-level effects such as metabolic remodeling, inhibition of proliferation, and induction of programmed cell death are often seen in the nanomolar to low micromolar range, depending on cell type, nutrient and stress conditions, and experimental setup. This molecule is commonly used to elucidate the dependence of tumor models (such as leukemia and brain tumor systems) on OXPHOS, evaluate metabolic vulnerabilities and synthetic lethality strategies (for example, in combination with inhibition of the glycolytic pathway), and support mechanism validation, target confirmation, biomarker exploration, and screening activities centered on mitochondrial respiration in in vitro cell models and mouse xenograft models; the concentrations or doses used in experiments typically depend on the specific experimental design and research objectives. For laboratory research use only. Not for human or veterinary use, diagnosis, or consumption.
