Fluticasone Furoate
Fluticasone furoate is a high-affinity synthetic glucocorticoid, commonly used in inhalation formulations for the treatment of asthma and allergic rhinitis. It is a GR receptor agonist; after binding to cytoplasmic GR, it translocates into the nucleus and binds to GRE to upregulate anti-inflammatory genes, downregulate the expression of inflammatory factors such as IL‑4, IL‑5, IL‑13, and TNF‑α, and inhibit the recruitment of inflammatory cells such as eosinophils and T cells. Its ester group structure increases lipophilicity and receptor affinity, providing a long local duration of action and enabling once-daily dosing; systemic exposure is low, but long-term use at high doses still requires monitoring for HPA axis suppression and local fungal infections.
