Background

FDI-6 (CAS No.: 313380-27-7) is a synthetic small-molecule transcription factor inhibitor and chemical biology probe designed for early-stage discovery research in gene regulation and oncology, primarily used to dissect transcriptional networks centered on Forkhead box protein M1 (FOXM1) and their associations with phenotypes. Studies have shown that it can directly bind to the FOXM1 protein, block its binding to DNA, and displace it from target-site chromatin, thereby selectively downregulating FOXM1-activated gene transcription; among homologous family members and related transcription factors, it exhibits functional selectivity for FOXM1, with advantageous inhibitory characteristics compared with FOXA1, FOXA2, FOXP2, GATA-1, and others. The resulting remodeling of transcriptional programs can perturb key proliferation-related pathways, including cell-cycle regulation, mitotic progression, DNA damage response, and stress adaptation. In in vitro experiments, FDI-6 typically shows inhibitory effects in the low tens of micromolar range on FOXM1 DNA-binding activity and FOXM1-dependent cellular phenotypes, while responses in cell proliferation and transcriptional readout endpoints are often observed from the low micromolar to low tens of micromolar range, consistent with its characteristics as a tool compound for mechanism validation and pathway analysis. Typical applications include target validation, transcriptional network mapping, mechanistic pathway dissection, and phenotypic screening, and it is commonly used in adherent cell systems (such as MCF-7 and MDA-MB-231 breast-derived models and the PEO1 ovarian-derived model) to evaluate FOXM1-dependent phenotypes and changes in downstream gene expression; the concentration or dose used in experiments usually depends on the specific experimental design and research objective.
