EHop-016
EHop-016 (CAS 1380432-32-5) is a small-molecule inhibitor targeting the Rac GTPases Rac1 and Rac3. It was designed to inhibit Rac activity, thereby modulating processes such as lamellipodia formation and cell migration.
EHop-016 mainly exerts its biological activity by disrupting the interactions between Rac and Vav2 as well as downstream effector molecules (such as PAK1). In cell-based studies, EHop-016 shows selective inhibition of Rac1, with an IC50 of 1.1 μM in MDA-MB-435 and MDA-MB-231 cells.
EHop-016 has potential applications in studying Rac-mediated cancer cell migration and metastasis.

Rac Activity Assays |
Rac activity is determined from lysates of the MDA-MB-435 and MDA-MB-231 human metastatic cancer cell lines. Cancer cells in culture medium (DMEM, 10% FBS, pH 7.5) are treated with vehicle (0.1% DMSO) or varying concentrations of EHop-016 (0–10 μM) for 24 h. Rac1 activity is determined using the G-LISA Rac1 activation assay kit. |

EHop-016 inhibited Rac activity and reduced Rac-directed lamellipodia formation in both MDA-MB-435 and MDA-MB-231 metastatic cancer cells. Moreover, EHop-016 decreased Rac downstream effects of PAK1 (p21-activated kinase 1) activity and directed metastatic cancer cells migration. |
