Background

D-(+)-Raffinose (hydrate) (CAS No.: 17629-30-0) is a naturally occurring trisaccharide widely distributed in plants, consisting of galactose, glucose, and fructose, and is commonly utilized in biomedical research as a functional carbohydrate with regulatory effects on metabolic and signaling processes. It is hydrolyzed by α-galactosidase into galactose and sucrose, linking it to carbohydrate metabolism and energy homeostasis pathways. Experimental evidence indicates that raffinose derivatives can modulate biological activity by interfering with specific enzymes and signaling pathways, including inhibition of lectin LecA and glucosyltransferase GtfC at low micromolar levels, as well as modulation of cyclic diguanylate signaling through enhanced phosphodiesterase activity. These effects contribute to reduced biofilm formation in bacterial systems such as Pseudomonas aeruginosa and Streptococcus mutans, highlighting its relevance in antimicrobial and anti-biofilm research. Additionally, raffinose has been shown to influence immune-related pathways by decreasing IL-4 and IL-5 mRNA expression, supporting its utility in studies of allergic inflammation and airway eosinophilia. In disease models, it is frequently employed to investigate metabolic dysregulation and carbohydrate-mediated signaling, as well as to explore potential therapeutic strategies targeting microbiological or immunological processes. The concentrations or doses used in experiments typically depend on specific experimental designs and research objectives, particularly in cell-based assays, microbial cultures, and in vivo models.
