Calcein AM (Calcein acetoxymethyl ester; CAS: 148504-34-1) is a non-fluorescent, cell-permeable derivative of calcein employed for viability assays and live-cell fluorescence staining. Upon entering live cells, intracellular esterases hydrolyze Calcein AM to release highly fluorescent calcein, a charged molecule unable to diffuse across intact membranes, resulting in intracellular retention and pronounced green fluorescence emission. Its negligible cytotoxicity and insensitivity to pH variations support broad use in mammalian viability assays, typically combined with propidium iodide (PI) for differentiating living versus compromised cells.
