Benzamide
Benzamide (CAS No.: 55-21-0) is a small aromatic amide and a classical inhibitor of poly(ADP-ribose) polymerase (PARP), widely utilized in biochemical and cellular studies investigating DNA damage response and repair pathways. By inhibiting PARP enzymatic activity, Benzamide modulates poly(ADP-ribosyl)ation processes that are critical for maintaining genomic stability and coordinating cellular responses to DNA strand breaks. In vitro studies demonstrate that Benzamide exhibits protective effects against glutamate- and methamphetamine-induced neurotoxicity, suggesting its utility in modeling oxidative stress and excitotoxic injury mechanisms, typically with activity observed in the low micromolar range. In animal research models, Benzamide has been shown to attenuate methamphetamine-induced dopamine depletion, supporting its role in studies of dopaminergic system integrity and neurochemical regulation, while not acutely altering striatal dopamine metabolism or thermoregulation. As a derivative of benzoic acid, it is frequently employed as a reference compound in PARP-related assays, neuroprotection studies, and screening platforms aimed at evaluating modulators of DNA repair and neuronal stress pathways, with experimental concentrations or dosing regimens adjusted according to specific study designs and research objectives.
