Background

Azide-PEG1-Val-Cit-PABC-OH (CAS 2055041-40-0) is a key cleavable linker for ADC (antibody-drug conjugate) development. Mechanistically, it enables efficient and stable conjugation through a copper-free click reaction between the azide group and DBCO-modified biomolecules. The PEG1 spacer optimizes both water solubility and steric hindrance. After the ADC is internalized by tumor cells, the Val-Cit dipeptide (product number: BA2571) sequence is specifically cleaved by lysosomal cathepsin B, followed by self-immolative dissociation of the PABC group to release the drug payload. The core functions focus on targeted delivery and controlled drug release. This linker is applied in the construction of tumor-targeted ADCs, bioorthogonal labeling and in vivo imaging, and the modification of nanodrug delivery systems. It also features excellent biocompatibility and plasma stability.
