Background

ATN-161 (CAS No.: 262438-43-7) is a synthetic pentapeptide derived from the synergy region of fibronectin and is classified as an integrin-targeting research peptide. It is mainly used in biomedical research and drug discovery for mechanistic studies related to cell adhesion, angiogenesis, and the tumor microenvironment. Its mechanism of action is characterized by competitive antagonism of integrin α5β1, and it can interact with other integrin subtypes, including αvβ3, thereby disrupting the binding of fibronectin to integrins, weakening integrin-dependent downstream signal transduction, and subsequently modulating endothelial and tumor cell migration, invasion, and phenotypes associated with neovascularization; in endothelial and tumor cell models, ATN-161 can inhibit VEGF-driven migration of human endothelial cells and multiple angiogenesis readouts, reflecting its blocking effect on the crosstalk between integrins and pro-angiogenic pathways; in addition, exploratory virology studies have also evaluated its ability to interfere with the binding of spike protein to α5β1, for mechanistic investigation of host factor interactions and not involving clinical use.
