Cy3.5 maleimide can be used for selective labeling of free sulfhydryl groups (e.g. cysteine residues in peptides, proteins and oligonucleotides). Before labeling, cystines should be prepared with reductant such as TCEP (tris-carboxyethylphosphine). The mono-reactive malemides is particularly good for the labeling of antibody, that’s because the dye molecules are located away from the antibody binding site and functioning of the labeled molecules can be protected.
