Nectin-4 is a cell adhesion molecule with limited expression in normal tissues, but it is highly overexpressed in various epithelial cancers-especially urothelial carcinoma-and participates in tumor cell proliferation, migration, and angiogenesis. Enfortumab is a humanized anti-Nectin-4 antibody that can be conjugated with the highly potent microtubule-disrupting agent MMAE to form the antibody-drug conjugate (ADC) Enfortumab vedotin-ejfv (development code F1132). Currently, Enfortumab is being investigated for the treatment of locally advanced and metastatic urothelial carcinoma.
