Aceglutamide
Aceglutamide (CAS No.: 2490-97-3) is an acetylated derivative of the conditionally essential amino acid L-glutamine, classified within glutamine and its functional derivatives, and is widely investigated as a neuroprotective small molecule in biomedical research. Owing to its ability to penetrate the blood–brain barrier, Aceglutamide has been extensively studied in the context of central nervous system disorders, particularly ischemic stroke and neurodegeneration. Mechanistically, it modulates multiple cellular defense pathways by enhancing antioxidant systems, including glutathione, thioredoxin, and Nrf2 signaling, while concurrently suppressing stress-associated mediators such as apoptosis signal-regulating kinase 1 and TRAF1. It also activates pro-survival signaling through the Akt/Bcl-2 axis, thereby reducing oxidative damage and inhibiting neuronal apoptosis, with notable effects observed in dopaminergic neurons of the substantia nigra. In preclinical models of cerebral ischemia and reperfusion injury, Aceglutamide has been shown to reduce infarct volume, improve neurological and motor function, and mitigate neuronal loss, supporting its utility in studying ischemia-related pathophysiology. In vitro and in vivo activities are generally evaluated across low micromolar to higher concentration ranges depending on the model system, with dosing regimens tailored to experimental design. Beyond neuroprotection, its role as a stabilized glutamine source has also supported investigations in metabolic and nutritional models. Collectively, Aceglutamide serves as a versatile tool compound for probing oxidative stress responses, apoptosis regulation, and neuroprotective mechanisms in drug discovery and translational neuroscience research.
