5(S)-HETE (CAS 70608-72-9) is a bioactive lipid mediator generated from arachidonic acid via the catalytic action of 5-lipoxygenase (5-LOX). Mechanistically, it exerts its effects by specifically binding to receptors such as BLT2 and GPR99, thereby activating downstream signaling pathways including PI3K/Akt and MAPK. This regulates the release of inflammatory cytokines, chemotaxis of immune cells, as well as cell proliferation and apoptosis. Bioactivity data show that its chemotactic EC50 for human neutrophils is approximately 10–100 nM, its binding Ki value for the BLT2 receptor is about 35 nM, and in asthma models, it can induce airway smooth muscle contraction (EC50 approximately 200 nM).
