4-Hydroxybenzyl alcohol
4-Hydroxybenzyl alcohol (CAS No.: 623-05-2) is a naturally occurring phenolic compound widely distributed in plants, including species of the genus Gastrodia, and represents the aglycone form of gastrodin with diverse pharmacological activities relevant to biomedical research. It is primarily characterized as a bioactive small molecule with anti-inflammatory, antioxidant, neuroprotective, and anti-nociceptive properties, and has been extensively investigated in the context of neurodegenerative disorders, inflammation, and oncology. Mechanistically, 4-hydroxybenzyl alcohol modulates key cellular processes by suppressing lipopolysaccharide-induced production of nitric oxide and reactive oxygen species, downregulating inducible nitric oxide synthase expression without significantly affecting cyclooxygenase-2, and attenuating oxidative stress–mediated signaling pathways. In vitro studies demonstrate activity in the low millimolar to high micromolar range in macrophage and neuronal cell systems, while in vivo models show efficacy in reducing inflammatory exudate formation, leukocyte infiltration, nociceptive responses, and ischemia-induced neuronal damage, as well as inhibiting tumor angiogenesis and growth. Additionally, behavioral studies indicate anxiolytic-like effects, further supporting its neuromodulatory potential. Owing to these multifunctional biological effects, 4-hydroxybenzyl alcohol is commonly utilized in cell-based assays and animal models to investigate mechanisms of inflammation, oxidative stress, and neuroprotection, as well as in early-stage drug discovery efforts targeting central nervous system disorders and cancer, with experimental concentrations or dosing regimens adjusted according to specific study designs.
