2',3'-cGMP sodium
2',3'-cGMP sodium(CAS No.: 15718-49-7)is the sodium salt form of 2',3'-cGMP. 2',3'-cGMP is an endogenous cyclic nucleotide in mammals, mainly derived from RNA degradation, metabolized by 2',3'-cyclic nucleotide 3'-phosphodiesterase(CNPase)to 2'-GMP/3'-GMP and ultimately to guanosine, constituting the 2',3'-cGMP - guanosine pathway; it can competitively inhibit the high-affinity cGMP transporter in erythrocytes and weakly activate cGMP-specific ATPase; the pathway end product guanosine has neuroprotective, renoprotective, and anti-inflammatory and antioxidant activities; only when generated intracellularly can it participate in the apoptotic process by regulating mitochondrial pore opening, while exogenous administration has no direct apoptosis-inducing activity.
Human erythrocyte inside-out vesicle experiment: after co-incubation with 1 μM [³H] 3',5'-cGMP at 37℃ for 120 min, the Ki for inhibition of its high-affinity transport was 4.1±0.3 μM; at a concentration of 2 μM, it increased cGMP-specific ATPase activity to 430±160% of the basal value. When applied exogenously to A549 lung adenocarcinoma cells, it did not induce apoptosis.
In anesthetized mice, infusion via the jugular vein at a rate of 0.3 μmol・kg⁻¹・min⁻¹ for 45 min significantly increased urinary excretion of 2',3'-cGMP, 2'-GMP, 3'-GMP, and guanosine; basal urinary 2',3'-cGMP excretion was increased by 147% in CNPase knockout mice; this infusion dose had no significant effect on mean arterial pressure in mice.
