HPF(Hydroxyphenyl fluorescein) is a cell-permeable aromatic amino-fluorescein derivative that has little intrinsic fluorescence.HPF reacts with intracellular peroxynitrite through hydroxyl radicals, generating intense green fluorescence. It offers high specificity and stability: oxidation occurs only in the presence of highly reactive oxygen species (hROS) such as hydroxyl radicals, peroxynitrite, and hROS generated by peroxidase/H₂O₂ systems. HPF does not react with hypochlorite ions, nitric oxide, hydrogen peroxide, superoxide, or other oxidants. Upon oxidation, HPF converts to the highly fluorescent molecule fluorescein, with excitation/emission peaks at 490/515 nm, making the direct detection of highly reactive biological radicals simple and intuitive. HPF can be used in fluorescence microscopy, high-throughput imaging systems, fluorescence plate readers, or flow cytometry.
