HBC (HBC 530) is a synthetic dye similar to GFP fluorophores, featuring a structurally rigid electron acceptor and strong electron donor structure. HBC itself does not fluoresce in solution, but when it binds to Pepper (RNA aptamer), it forms a tight complex, activates and emits bright fluorescence, with a binding constant (Kd) of approximately 3.5 nM. The emission peaks of HBC vary in different Pepper complexes, covering a visible spectrum ranging from cyan to red. The typical excitation/emission wavelength of HBC 530 is 485/530 nm, which is highly suitable for biological imaging applications such as live cell imaging of RNA.
