Background

WM-1119 (CAS No.: 2055397-28-7) is a synthetic small-molecule chemical probe for discovery research in epigenetics and cancer biology, selectively targeting the lysine acetyltransferases KAT6A (MOZ) and KAT6B (MORF), and is used to dissect molecular programs associated with mammalian cell lineage determination and proliferation by interfering with histone acetylation and chromatin-mediated transcriptional regulation; its binding affinity for KAT6A is in the nanomolar range (Kd approximately 2 nM), markedly stronger than its binding to the related acetyltransferases KAT7 (HBO1) and KAT5 (TIP60) (Kd approximately 0.5 μM and 2.2 μM, respectively), and it shows minimal activity across a broad off-target panel under micromolar screening conditions; in in vitro enzymatic assays, it exhibits low-nanomolar inhibitory activity (approximately 6.3 nM), while in cellular models such as B-cell lymphoma, it shows antiproliferative effects in the submicromolar range (IC50 approximately 0.25 μM), highly consistent with efficient occupancy of the target acetyltransferases and regulation of chromatin function; this chemical probe is commonly used to define KAT6A/KAT6B-dependent pathways, perform target validation, support screening cascades and structure–activity relationship studies, and evaluate epigenetic readouts, pharmacodynamic markers, and related phenotypic responses in disease-relevant cellular systems and mouse tumor research models; the concentrations or doses used in experiments are typically determined according to the specific research objectives and experimental design.
