Trolox

Trolox (CAS No.: 53188-07-1) is a water-soluble, cell-permeable analogue of vitamin E widely utilized in biomedical research as a reference antioxidant and positive control in assays evaluating oxidative stress and free radical scavenging capacity. Functionally classified as a small-molecule antioxidant, Trolox acts primarily by neutralizing reactive oxygen species and inhibiting lipid peroxidation, thereby protecting cellular membranes and macromolecules from oxidative damage. Mechanistically, it modulates redox-sensitive signaling pathways associated with apoptosis and cellular survival, including attenuation of oxidative stress–induced DNA fragmentation and regulation of pro- and anti-apoptotic protein expression. In vitro, Trolox demonstrates protective effects against hydrogen peroxide–induced cytotoxicity and apoptosis in the low micromolar range, though effective concentrations vary depending on cell type and experimental conditions. It is extensively applied in studies of oxidative injury, aging, neurodegeneration, and cancer biology, as well as in high-throughput screening platforms assessing antioxidant capacity. In vivo, Trolox has been used in animal models to mitigate ischemia-reperfusion injury and support neuronal survival, with dosing regimens tailored to specific experimental designs. Its well-characterized antioxidant profile and reproducible activity make it a valuable tool compound in drug discovery and mechanistic studies focused on oxidative stress–related pathologies.
