MitoTEMPOL
MitoTEMPOL (CAS No.: 1101113-39-6) is a mitochondria-targeted nitroxide and superoxide dismutase mimetic widely utilized in redox biology and mitochondrial research, designed by conjugating the antioxidant moiety TEMPOL to a lipophilic triphenylphosphonium cation that facilitates selective accumulation within the mitochondrial matrix. Functionally, MitoTEMPOL catalytically converts mitochondrial superoxide anion to hydrogen peroxide, thereby modulating intracellular reactive oxygen species homeostasis and enabling mechanistic investigation of oxidative signaling pathways, including those governing mitochondrial respiration, apoptosis, and redox-sensitive transcriptional regulation. As a targeted antioxidant, it is commonly applied to dissect the role of mitochondrial oxidative stress in pathological contexts such as neurodegeneration, cardiovascular dysfunction, metabolic disorders, and inflammation. In vitro, MitoTEMPOL exhibits activity in the low micromolar to micromolar range depending on cell type and oxidative conditions, reflecting its efficiency in scavenging mitochondrial superoxide and influencing downstream redox signaling. In experimental settings, it is broadly employed in cultured cells and animal models to probe mitochondrial function, validate oxidative stress–related targets, and support drug discovery efforts aimed at modulating mitochondrial ROS, with effective concentrations or dosing regimens varying according to specific study designs and biological endpoints.
